SUMO化修饰与早幼粒白血病蛋白核体形成 *

摘要 早幼粒白血病蛋白核体(promyelocytic leukaemia nuclear bodies,PML NBs)是哺乳动物细胞中普遍存在的一种亚核结
构,广泛参与如转录调节、基因组稳定性维持、抗病毒、细胞凋亡、肿瘤抑制等一系列的生物学事件.SUMO(small
ubiquitin modifier)修饰是蛋白质翻译后修饰领域中的研究热点,SUMO修饰对 PML核体的形成与降解都发挥着重要作用.
近年来研究发现,人的 E3泛素连接酶 RNF4(RING finger protein 4),可促进依赖 SUMO-2/3修饰的 PML核体的泛素化连接,
并且 ATO(三氧化二砷)可加速其对 PML核体的降解.荧光共振能量转移(fluorescence resonance energy transfer,FRET)技术可
完全应用于活细胞内 PML核体和 SUMO蛋白之间在时间和空间上的精确互作.因此,更深入地研究 PML核体形成和降解
的机理以及在这个过程中重要蛋白质之间的相互作用具有重要而深远的意义.

1991年,De Th佴 等首先发现染色体 t(15, 17)
易位导致早幼粒白血病蛋白(PML)基因和 RAR琢(维
甲酸受体 琢)基因的融合从而引发白血病[1],随后对
PML 核体 (promyelocytic leukaemia nuclear bodies,
PML NBs)结构和功能的研究发现,其在广泛的生
物学活动中发挥重要作用,为更深入地研究其细胞
学功能提供了重要依据.虽然目前对于定位在
PML核体上的蛋白质已有大量研究,但对于 PML
核体如何形成、降解及该过程发生的精确亚细胞位
置还不清楚.本文主要
SUMO(small ubiquitin
modifier)修饰在 PML 核体形成及降解中的研究
进展.
1 PML核体简介
PML基因 N端包含保守的 TRIM或者 RBCC
结构,由一个环指结构(RING finger domain)、一个
或两个 B-Box结构和一个卷曲螺旋结构域(coiled-
coil domain)[2-3]组成,PML蛋白的结构对于 PML核
体的形成非常重要,虽然 C端由于可变剪切可产
生至少 11个蛋白异构体,但已证明多达 6种 PML
异构体(玉~遇)均能形成 PML核体[4].
PML核体又称 Kremer小体、核域 -10(ND-10)
和 PML致瘤域(PODs)[1],是染色质之间的点状核
斑结构,广泛存在于大多数哺乳动物细胞与组织
中,根据细胞类型、细胞周期阶段的不同,平均每
个细胞核质中约有 1~30 个核体,直径为 0.2~
10 滋m[5-6].作为蛋白质性质的结构,PML核体是和
核质紧密联系在一起的,电子显微镜下可观察到它
包含一个环指状的蛋白质结构域并且中心没有检测
到有核苷酸[5-6].本实验室用电子显微镜和免疫荧
光方法观察到 PML核体在细胞中的分布(图 1).
PML核体是一种动态的结构,不管是在正常
情况下还是在细胞胁迫下均在数量、大小和位置上
发生巨大的变化[5-9],活细胞成像
显示,虽然
大多数期间的 PML核体呈稳定状态,大约 12%的
PML核体
现出依赖 ATP的变化[9-11].并且 PML
蛋白定位或者离开 PML核体都会直接影响 PML核
体的结构.

在细胞周期中,PML核体不仅在数量上而且
在生化组成上发生巨大变化[12].早期的研究表明,
G0期细胞中几乎没有 PML核体,当细胞进入 G1
并移入 S和 G2期,PML核体数量增加[13].在 S期
早期,PML核体依然和染色质紧密联结,因为染
色质的修饰失去它们的结构整合性,进而分解形成
小体.G2期的数量相当于 G1期的大约 2倍[14].分
裂期随着染色质的固缩,PML核体数量减少,形
成更大的聚集体 MAPPs(PML蛋白有丝分裂积累
物)并和中期染色体分离,然而有一部分仍然呈联
合状态,此时的 MAPPs成为子细胞核 PML蛋白
的来源[15].除了 M期,SUMO蛋白和 SP100蛋白
都是 PML核体的组分,Daxx蛋白在 S期的晚期由
于定位在异染色质上和 PML核体分离,G2期的
Daxx蛋白数量少于 G1期[6].
2 SUMO化修饰与 PML核体形成的关系
目前已经证实长期或暂时定位在 PML核体中
的蛋白质超过 50 个,如 sp100、Daxx、SUMO
等[7-8],这些蛋白质直接或间接地参与 PML核体重
要生物学功能的发挥.对于 PML核体.直至今日
我们仍不清楚在细胞核中它的形成过程以及其发生
的具体位置,但目前的研究结果表明 SUMO修饰
在 PML核体的形成中具有重要的作用[6, 16-17].
SUMO化修饰是指 SUMO蛋白共价结合于靶
蛋白的赖氨酸残基上,这个过程类似但又不同于泛
素化,而且与泛素介导蛋白质的降解不同 [18].
SUMO 化修饰是一种重要的蛋白质翻译后修饰,
在蛋白质定位、染色质结构的维持、转录调节、
DNA 损伤修复、核质运输、信号转导等方面发
挥作用 [19-22]. 已知人类的 SUMO 基因有 4 个:
SUMO-2、3、4之间有 83%以上的同源性,但它们
和 SUMO-1 的同源性低于 48% [23].SUMO 化修饰
靶蛋白的过程是:a.成熟.SUMO前体蛋白在蛋
白酶的作用下成熟,暴露出 C端 Gly.b.激活.
ATP水解提供能量使 SUMO的 C端 Gly与 E1-激
活酶(SAE-1,SAE-2)的一个 Cys残基通过硫酯键
相连接. c.缀合.激活的 SUMO 经 E1 传递到
E2- 结合酶(Ubc9),与 E2 活性位点的 Cys93 残基
借助硫酯键相连接.d.连接.SUMO的 Gly残基
与靶蛋白 Lys 残基的 着2 氨基通过异肽键结合.
Rodriguez等[24]证明,SUMO的靶蛋白含有共有序
列“追KXE”及核定位信号 (nuclear localization
signal,NLS),其中 追为疏水性氨基酸残基,K为
Lys,X为任意氨基酸,E为 Glu. e.通过异肽键
酶的作用,SUMO与靶蛋白分离,回到初始状态,
称为去 SUMO 化( desumoylation).去 SUMO 化则
由 SUMO化的特异性蛋白酶(SENP)催化,它们定
位于不同的亚细胞区[19-20].
研究发现,PML 蛋白上 3 个位点 (65、160、
490)分别突变后不被 SUMO化修饰,Zhong等[17]发
现,向 PML-/-细胞中转染不能被 SUMO 修饰的
PML突变体,不能形成核体,说明这 3个位点都
是 PML核体形成及其完整性维持必需的.特别是
PML的 K160位与 SUMO-2/3共价结合时,对相关
蛋白募集到核体是很关键的.体外实验已经证明,
SUMO二类家族都可以修饰 PML蛋白,而且这种
修饰以不同于 SUMO-1作用的机理对 PML核体的
形成造成影响[25-29].表现在修饰位点上:PML蛋白
的 490位点主要被 SUMO-1修饰,160位点主要被
SUMO-2/3修饰并且形成多 SUMO链修饰(图 2).
Pandolfi等的研究发现 [30-31]:SUMO 化修饰过
程中的某些组分也影响 PML核体的形成,体外培
养的细胞中缺失 Ubc9可导致 PML核体结构和功
能的严重缺陷,并且 PML蛋白不能被 SUMO化修
饰,引发 PML核体裂解;过量表达的 SuPr1(一种
SUMO蛋白酶)使 PML核体形成为数很少的较大聚
集体,并且使定位在 PML核体的蛋白质(如 Daxx
蛋白和 CBP蛋白)发生重新分布,如 Daxx蛋白和
CBP 蛋白.这些酶的改变又为 SUMO 化修饰在
PML核体形成中扮演重要角色提供了有力证据.
Shen等[32-33]发现 PML上存在一个叫做 SUMO
蛋白结合序列(sumo interaction motif,SIM)的结构,

该结构和它的 SUMO化位点不同,使 PML蛋白能
够与 SUMO化修饰的蛋白质包括它自身发生非共
价结合,PML蛋白中的 SIM也能够与发生 SUMO
化修饰的 PML结合,多个 PML蛋白通过 SUMO
形成大的复合结构,而其他能被 SUMO化修饰的
蛋白质或具有 SIM的蛋白质也能定位到这个复合物
中[34-35],因此 PML的形成依赖于被 SUMO 修饰的
SUMO 蛋白和 PML的多个 SIM 的非共价相互作
用[36].但最近有研究者向 PML-/-细胞中转染不含
SIM 的 GFP- PML 遇时,也形成了 PML 核体 [4],
因此推断 SIM也许在 PML核体形成中发挥重要作
用.对于 SIM是否是 PML核体形成的必需结构,
目前还没有达成共识,需要更确凿的实验论证.
3 人 E3泛素连接酶 RNF4及三氧化二砷诱
导对 PML核体的降解作用
早期的研究发现三氧化二砷(ATO)本身可能治
疗急性早幼粒细胞白血病(APL)[37],后来 De Th佴教
授等又证明砷特异性地降解 PML-RAR琢融合蛋白,
十余年的研究结果使 ATO降解 PML核体的机理日
渐明了.
在 PML核体降解过程中,人 E3泛素连接酶
(RING finger protein 4, RNF4)的参与和 SUMO修饰
是不可或缺的重要过程.其中,小核指环状蛋白
RNF4是一个人的 E3泛素连接酶,它和酵母中依
赖 SUMO的泛素连接酶同源,包含一个 C端的环
状指和一个对许多肿瘤抑制蛋白和 E3泛素连接酶
特异的基序.最近的研究发现,RNF4是第一个能
够特异结合多 SUMO 链的蛋白质 [38].作为 RNF4
忠实的目标,PML是第一个发现的被泛素降解但
要依赖 SUMO化修饰的蛋白质[39].
当没有砷诱导时, PML160 位点形成多链
SUMO-2/3,RNF4结合多 SUMO链后,形成多泛
素链,之后 PML核体被蛋白酶降解,401位点可
直接发生多泛素修饰[39],而 SUMO-1修饰 65位其
自身不能被泛素修饰,只可间接促进 160位点的多
SUMO链形成和直接稳定 RNF4的结合[40-41].当有
砷诱导时,ATO 引起 SUMO-2/3 链的大量增加,
PML核体降解过程被加速,而 PML 160位点突变、
RNF4的缺失或者蛋白酶抑制引起混合的、多泛素
化的、多 SUMO链的积累,导致砷不能诱导 PML
的降解和核内 SUMO修饰的 PML的积累[40, 42](图 3).

Percherancier 等 [43]用生物发光共振能量转移
(BRET)方法发现,PML的 SIM/SBD(SUMO binding
domain)在 ATO-RNF4 诱导的 PML核体降解途径
中必不可少,而 ATO的增强不是必需的,并且在
核体中 SUMO 修饰和依赖 SIM的 PML聚集作用
足够引起 RNF4介导 PML核体降解.最近,我们
用活细胞荧光共振能量转移 (FRET)实验证明
ATO-RNF4通过多聚 SUMO-2修饰介导 PML核体
降解 (结果未发表).
4 荧光共振能量转移 ( FRET) 技术在研究
PML核体分子动态变化中的应用
近年来,随着水母中绿色荧光蛋白及珊瑚和海
葵中红荧光蛋白的克隆并在各种异源细胞中的表
达,绿色荧光蛋白(GFP)作为一种新型的探针分子,
利用其研究活细胞内蛋白质空间分布和相互作用成
为可能[44-45].比如有研究者利用光漂白后荧光恢复
技 术 (fluorescence recovery after photobleaching,
FRAP),观察发现,大多数情况下 GFP-PML 和
GFP-SP100蛋白在 PML核体是稳定的,当 PML蛋
白从核质移动到 PML核体外,PML核体也随之发
生变化[46].
GFP-FRET结合图形显微技术和计算机处理技
术,为活体细胞内蛋白质动力学研究提供了一种可
视化的分析方法[47].GFP-FRET图像显微术结合荧
光寿命显微图像,已被广泛应用于基因转染、表达
病毒感染致病机理研究、蛋白质定位和蛋白质动力
学研究、蛋白质定量分析、蛋白质运输荧光共振能
量转移、光漂白荧光关联谱学、细胞分类选择和细
胞系分析等[48].
已有研究者运用 FRET技术研究了 PML核体
和 SUMO 蛋白之间在时间和空间上的精确互作,
并比较 SUMO-1和 SUMO-2/3对 PML核体修饰的
差异.因此,此项研究可完全应用于为全面揭示
SUMO 蛋白对 PML 核体的修饰作用,进而发现
其对 PML核体形成及降解的影响提供重要的研究
方法.
5 展 望
目前,虽然定位于 PML核体上蛋白质研究得
较多并且在鉴别及功能研究方面有较大的发展,但
是还存在一系列因素制约了 PML核体机理的研究:
a.还未能用生物化学的方法提纯 PML核体;b.
存在许多 PML蛋白异构体;c.现在还不清楚哪些
成分在同一时间保持不变;d.鉴定出来的成分到
底是核质还是核体的 PML伙伴.近年来,随着分
子影像技术的发展和绿荧光蛋白的广泛应用,荧光
共振能量转移作为唯一能在生理条件下反映活细胞
体内蛋白质相互作用的方法,为开展和 PML核体
的形态建成、功能有关的研究,特别是 SUMO家
族对 PML核体的修饰作用的研究奠定了基础.

SUMOylation and PML NBs Formation*

Abstract PML NBs (promyelocytic leukaemia nuclear bodies) is subnuclear structure in mammalian cells, which
widely takes part in kinds of biological events such as transcriptional regulation, genome stability, response to viral
infection, apoptosis and tumor suppression. As protein post-translational modification, SUMO located in nucleus
plays important roles in PML NBs formation and degradation. Recent research suggests that the human E3
ubiquitin ligase-RNF4 (RING finger protein 4), mediates PML NBs degradation depending on SUMO-2/3
modification of PML, ATO(arsenic trioxide) could facilitate in this process. FRET(fluorescence resonance energy
transfer)technology could be used in studying the protein-protein interaction of PML NBs and SUMO protein in
living cells. Thus, it is far-reaching and significant to research deeply in the mechanism of the formation and
degradation of PML NBs and the interaction between the important proteins during this pathway.
