沙葱多糖对小鼠腹腔巨噬细胞的激活作用及抗氧化功能的研究 附件2 中英文摘要格式 作者姓名:王丽思 论文题目:沙葱多糖对小鼠腹腔巨噬细胞的激活作用及抗氧化功能的研究 作者简介:王丽思,女,1984年2月出生,2007年9月师从于内蒙古农业大学敖长金教授,于2010年6月获得农学硕士学位。 中 文 摘 要 本论文研究了沙葱多糖对小鼠腹腔巨噬细胞的激活作用及其抗氧化功能。主要研究与结论如下: 1.采用中性红吞噬法、MTT法、ELISA法检测了沙葱多糖浓度分别为50、100、200、400、 -1800μg.ml对小鼠腹腔巨噬细胞吞噬中性红的能力、能量代谢水平及IL-6分泌的影响。结果 -1表明,当沙葱多糖浓度为100μg.ml时,可视为对巨噬细胞激活的最适浓度。 -12.采用上述方法,研究沙葱多糖在最适浓度100μg.ml时,比较0h、12h、24h、 36h、48h后对小鼠腹腔巨噬细胞的激活作用。研究表明,24h时该浓度下的沙葱多糖对小鼠腹腔巨噬细胞的激活作用较好。 ,3.通过体外抗氧化试验法研究了沙葱多糖清除O?和抑制?OH的能力。结果表明,沙2 ,葱多糖具有清除O?和抑制?OH的能力,浓度越大抗氧化作用越强。其中,5mg/mL的沙葱2 ,多糖清除O?的能力最强;1 mg/mL的沙葱多糖抑制?OH的能力最强。 2 4.本试验将50只小鼠随机分为5组,每组10只。每天给小鼠灌胃空白对照组0.9%生理 -1盐水、阳性对照组30mg?kgVE、沙葱多糖高、中、低剂量组分别为800、400、200mg/kg?BW。预饲期7d,正式试验期15d后眼球采血并分离血清,用试剂盒测定小鼠血清及肝、肾、心组织中的总超氧化物歧化酶(T-SOD)、丙二醛(MDA)、过氧化氢酶(CAT)、总抗氧化能力(T-AOC)、谷胱甘肽过氧化物酶(GSH-Px),研究沙葱多糖对小鼠血清及各组织抗氧化能力的影响。结果表明,沙葱多糖能够增强小鼠血清、肝脏、肾脏和心脏中T-SOD、CAT 、GSH-PX活力及T-AOC,并减少MDA含量。与空白对照组相比,?血清和肝脏中,沙葱多糖800mg/kg?BW 的T-SOD活力最强(P,0.05);在肾脏和心脏中,沙葱多糖400mg/kg?BW的T-SOD活力最高(P,0.05)。?血清和组织中,沙葱多糖400mg/kg?BW降低MDA的效果最好,除心脏差异不显著(P,0.05)外,其余差异均显著(P,0.05)。?血清和肝脏中,沙葱多糖400mg/kg?BW提高CAT活力的效果最好(P,0.05);在肾脏和心脏中,沙葱多糖800mg/kg?BW的效果最强(P,0.05)。在肾脏中CAT活力差异显著,在心脏中差异不显著(P,0.05)。?在血清和心脏中,沙葱多糖800mg/kg?BW提高T-AOC的作用效果最好(P,0.05);在肝脏中,沙葱多糖200mg/kg?BW的作用效果最好(P,0.05);在肾脏中,沙葱多糖400mg/kg?BW的效果最好(P,0.05)。?在血清中,沙葱多糖800mg/kg?BW提高GSH-PX活力的作用效果最好(P,0.05);在肝脏中, 沙葱多糖200mg/kg?BW的GSH-PX活力最强(P,0.05);在肾脏和心脏中,沙葱多糖400mg/kg?BW 的作用效果最好(P,0.05)。综合考虑整个试验结果,沙葱多糖中剂量组400mg/kg?BW的体 内抗氧化作用效果最佳。 关键词:沙葱多糖;小鼠;巨噬细胞;抗氧化 Effects of Polysaccharides extracted from Allium Mongolium Regel on Activation of peritoneal Macrophages and Antioxydation in Mice Wang Lisi ABSTRACT This thesis was conducted to study the effect of polysaccharides extracted from Allium Mongolium Regel on activation of peritoneal macrophages and antioxygen in mice.The main contents and conclusions of paper were as follows: 1.The effect of Polysaccharides extracted from Allium Mongolium Regel levels (50 , 100 ,200 , -1400 , 800μg.ml )on the phagocytosis capability of the mouse peritoneal macrophages,energy metabolism level and IL-6 were studied by neutral red phagocytosis test, MTT method and ELISA method. The results indicated that the concentration of Allium Mongolicum Regel Polysaccharides -1was 100μg.ml, which was the best concentration for activation of peritoneal macrophages. -1 2. To determine after 0, 12, 24, 36, 48h, 100μg.ml ofthe best concentration of Allium Mongolicum Regel Polysaccharides on the activation of peritoneal macrophages was tested following the above methods. The results showed that the effect of polysaccharides extracted from Allium Mongolium Regel on activation of peritoneal macrophages at 24h was better.,3. O?and ?OH were cleaned up by polysaccharides extracted from Allium Mongolium Regel 2,in vitro antioxygen test. The results indicated that O?and ?OH were cleaned up by the 2 polysaccharides extracted from Allium Mongolium Regel, the antioxidation was increased with the concentration. 5mg/mL of polysaccharide extracted from Allium Mongolium Regel has the strongest ,ability to clean up O?; and 1mg/mL of polysaccharide extracted from Allium Mongolium Regel 2 has the ability to inhibit ?OH strongly. 4. In this experiment, 50 mice were randomly divided into 5 groups of 10mices. Blank control -1group was given 0.9% saline per day, positive control group was given 30mg •kgVE, different doses of Allium Mongolium Regel polysaccharide groups were 800, 400, 200 mg / kg • BW, respectively. After 15d of the formal trial period, blood was sampled from eyeball and also sampled the liver, kidney, and hart, to investigated the antioxydation effects of polysaccharides extracted from Allium Mongolium Regel on T-SOD, MDA, CAT, T-AOC, GSH-PX of serum, liver, kidney and heart in mice. The results indicated that polysaccharides extracted from Allium Mongolium Regel has a certain antioxidant effect on the body. Allium Mongolium Regel polysaccharides improved T-SOD, CAT, T-AOC, GSH-PX of serum, liver, kidney and heart and it could reduce the content of MDA. Compared with the blank control group,?In the serum and liver, T-SOD of 800mg/kg • BW of the Allium Mongolium Regel polysaccharides was the strongest (P,0.05); In the kidney and heart, T-SOD activity of 400mg/kg • BW of the Allium Mongolium Regel polysaccharides was the highest (P,0.05). ?In the serum and tissues 400mg/kg • BW of the Allium Mongolium Regel polysaccharides can be best to decrease MDA. In addition to the heart of the difference was not significant(P,0.05), the other differences were significant (P,0.05).?In the serum and liver, 400mg/kg • BW of the Allium Mongolium Regel polysaccharide was the best to increase CAT(P,0.05); In the kidney and heart, 800mg/kg • BW of the Allium Mongolium Regel polysaccharide was strongest. In kidney,CAT activity was significantly different (P,0.05), but the difference was not significant in heart (P,0.05). ? In the serum and heart, 800mg/kg • BW of the Allium Mongolium Regel polysaccharides had the best effect to increase the T-AOC(P,0.05); In liver, 200mg / kg • BW of the Allium Mongolium Regel polysaccharides had the best role(P,0.05); In kidney, 400mg/kg • BW of the Allium Mongolium Regel polysaccharides was the best(P,0.05). ? In serum, 800mg/kg • BW of the Allium Mongolium Regel polysaccharide efficiently enhanced GSH-PX activity (P,0.05); In liver, GSH-PX activity of 200mg/kg • BW of the Allium Mongolium Regel polysaccharide was highest (P,0.05); In kidney and heart, 400mg/kg • BW of the Allium Mongolium Regel polysaccharide improve GSH-PX activity efficiently(P,0.05). In conclusion, 400mg/kg • BW of the Allium Mongolium Regel polysaccharide goup had better antioxidation in vivo than others.
