野鸟蓝舌病毒带毒调查 野鳥藍舌病毒帶毒調查 疫學研究組 丁履紉 助理研究員 摘要 本試驗配合家禽流行性感冒候鳥帶毒監測計畫之樣材,應用於藍舌病抗原監測。2006年至2008年野鳥排遺之藍舌病病毒核酸檢出率分別為0.15 %,7/4,541,、0.34%,14/4,093,和0.38%,12/3,163,。檢出之鳥種以雁鴨科、鹬鴴科和鷺鷥科為主。 33個藍舌病病毒核酸陽性的野鳥排遺,僅自鷸鴴科排遺分離出2株藍舌病病毒命名為WB989和WB990,分析其L2基因部份序列之親緣關係與KM株核苷酸相似性100%,故野鳥分離株屬於第2血清型為台灣本土型。比較33個陽性野鳥排遺、臺灣KM株、PT株等共35株其NS3部份序列親緣關係,發現可以分類為2群,其中4株野鳥排遺與台灣分離株及其它野鳥排遺不同群,核苷酸相似性介於85.4-86.1%。 The Survey of Bluetongue virus in Wild Birds Lu-Jen Ting Abstract The aim of the project is cooperated with the avian influenza surveillance project to work on Bluetongue virus. The detection rates of BTV from fecal of wild birds samples from 2006-2008 using RT-PCR test were conducted, the results were 0.15% (7/4,541), 0.34% (14/4,093), 0.38% (12/3,163), respectively. The major positive bird species were Anatidae, Charadriiformes and Ardeidae. Only two BTV viruses were isolated from 33 RT-PCR positive samples. The full length of the L2 gene of these two isolates, named WB989 and WB990, were sequenced and compared for the phylogenetic association with the sequences of BTV isolates published in Genbank. Bird isolates clearly segregated to serotype 2 and it showed the closest genetic relationship KM strain with 100% nucleotide sequence homology. Sequence analysis of partial NS3 of these 33 positive fecal samples, KM strain and PT strain, total of 35 strains, yielded two clades, whereas 4 fecal samples only 85.4-86.1% similarity with Taiwanese strains.
